Glu (+) Gal + Glu (+) Gal + Bait is up-regulated/mutated to a high background of transcriptional activation. GAL1 promoter is slightly leaky; both proteins are very stable; interaction occurs with high affinity. Take one clone for confirmation of interaction; store the rest.
Glu − Gal + Glu − Gal − Yeast mutation occurred that favors growth or transcriptional activation on galactose medium. Some bait/interactor combinations are known to activate lacZ versus LEU2 preferentially, or vice versa. If all other candidates winnow out, check these clones.
All other phenotypes Contamination/plasmid rearrangements/mutations Something really new Trash.
