Cell disruption buffer

Reagent Quantity (for 500 mL) Final concentration
KCl (2 M) 2.5 mL 10 mM
MgCl2 (1 M) 0.75 mL 1.5 mM
Tris-Cl (1 M, pH 7.5) 10 mL 20 mM
H2O, RNase-free 486.75 mL --
Immediately before use, for each 10 mL of buffer needed, add 10 μL of DTT (dithiothreitol; 1 M) to give a final concentration of 1 mM. Discard any unused DTT-containing buffer.
Prepare all reagents used to make this buffer with RNase-free H2O.
