Formaldehyde (2% in PBS)

MATERIALS

Reagents

HCl (1 N)

Paraformaldehyde, electron microscopy (EM)-grade (Prill form, Electron Microscopy Sciences)

Phosphate-buffered saline (PBS)

Equipment

Bottle, Pyrex, with cap

Fume hood

Hot plate

Stir bar, magnetic

METHOD

  • 1. Add the appropriate amount of EM-grade paraformaldehyde to PBS (e.g., 1 g of paraformaldehyde per 50 mL of PBS) in a Pyrex bottle with a stir bar.

  • 2. Heat the mixture on a hot plate in a hood with stirring at 60°C to dissolve the aldehyde.

    Keep the bottle cap loosened so that pressure does not build up.

  • 3. Allow the solution to cool to 20°C, and adjust the pH to 7.4 with 1 N HCl.
