Buffer A for Yeast Extracts

Reagent Quantity (for 100 mL) Final concentration
KCl (3 m)   0.33 mL        10 mm
HEPES (1 m, pH 7.9 at 25°C)     1 mL 10 mm (pH 7.0 at 4°C)
MgCl2 (1 m)   0.15 mL        1.5 mm
DTT (1 m)   0.05 mL        0.5 mm
PMSF (50 mm)     1 mL        0.5 mm
Leupeptin (1 mg/mL)   0.1 mL        1 µg/mL
Pepstatin (1 mg/mL)   0.1 mL        1 µg/mL
H2O to 100 mL
Add protease inhibitors just before use. Store at 4°C. Do not keep buffer to which protease inhibitors have been added for >1 wk.
