Splicing Buffer for Yeast Extracts (5×)

Reagent Quantity (for 100 µL) Final concentration
Polyethylene glycol (PEG) 8000 (50%, w/v) (use highest quality) 30 µL   15%
KPO4 (0.6 m, pH 7.0) 50 µL 36 mm
Spermidine (100 mm) 10 µL 10 mm
MgCl2 (125 mm) 10 µL 12.5 mm
Take care to be accurate when measuring viscous solutions such as PEG. Make splicing buffer fresh to avoid precipitation of KPO4 on storage.
