Imaging Medium (MEM-AIR)

Reagent Quantity Final concentration
Dulbecco’s minimal essential medium (DMEM) with high glucose and without l-glutamine, sodium pyruvate, or phenol red (Life Technologies 31053028) 48 mL
Fetal veal serum (FVS)  5 mL 10% (v/v)
HEPES (1 m)  1 mL  20 mm
l-Glutamine (200 mm) (Life Technologies) 500 µL   2 mm
Sodium pyruvate (100 mm) (Life Technologies) 500 µL   1 mm
In cases of long observations, the medium can be supplemented with 1% (v/v) penicillin–streptomycin. Use a stock solution containing 10,000 units of penicillin (base) and 10,000 µg of streptomycin (base)/mL utilizing penicillin G (sodium salt) and streptomycin sulfate in 0.85% saline (Life Technologies).
