Fixation Buffer
| Reagent | Amount for 700 mL |

| Paraformaldehyde | 21 g |
| NaH2PO4·H2O | 2.2 g |
| NaHPO4 (anhydrous) | 11.5 g |
| Ultrapure H2O | to 700 mL |
| NaOH (10 n) | 2–3 drops |
| Heat solution to 60°C with continuous stirring until cleared; do not exceed 70°C. (Note that paraformaldehyde does not dissolve easily in water at 60°C. The addition of a few drops of 10 n NaOH helps to promote paraformaldehyde depolymerization, and the solution will then appear clear.) Prepare this solution fresh; it is sufficient to fix ∼240 slides. | |
