Fixation Buffer

Reagent Amount for 700 mL
Paraformaldehyde     21 g
NaH2PO4·H2O    2.2 g
NaHPO4 (anhydrous)   11.5 g
Ultrapure H2O to 700 mL
NaOH (10 n) 2–3 drops
Heat solution to 60°C with continuous stirring until cleared; do not exceed 70°C. (Note that paraformaldehyde does not dissolve easily in water at 60°C. The addition of a few drops of 10 n NaOH helps to promote paraformaldehyde depolymerization, and the solution will then appear clear.) Prepare this solution fresh; it is sufficient to fix ∼240 slides.
