Luria–Bertani (LB) Medium Plus Ampicillin

| Agar | 20 g |
| NaCl | 10 g |
| Tryptone | 10 g |
| Yeast extract | 5 g |
| Prepare the above-listed ingredients in 1 L of deionized water. Adjust the pH to 7.0 with 5 n NaOH. Autoclave for 20 min at 15 psi (1.05 kg/cm2). Cool to ∼60°C and add ampicillin (final concentration 120 µg/mL). Pour the medium into Petri dishes (∼25 mL per 100-mm plate). Store the LB plates at 4°C; they will keep for at least 4 mo. | |
