Intracellular Recording Solution

Reagent Concentration
EGTA 0.5 or 10 mm
HEPES    10 mm
MgCl2    2 mm
Mg-ATP    2 mm
N-methyl-d-glucamine   140 mm
Use solutions containing 0.5 mm EGTA to record Ca2+-dependent inactivation (CDI) in Cav2 channels. Use solutions containing 0.5 or 10 mm EGTA to record the CDI of Cav2 or Cav1 channels, respectively, and 10 mm EGTA to record the CDF of Cav2.1 channels (high concentrations of EGTA inhibit the CDI of Cav2 channels). Adjust pH to 7.3 using methanesulfonic acid. (The osmolarity should be ~290–310.) Filter-sterilize. Store aliquots at −20°C.
