Synthetic Dextrose (SD) Plates without Ammonium Sulfate or Amino Acids (with or without Acetamide or Cytosine)
Mix yeast nitrogen base without ammonium sulfate and without amino acids (1.7 g/L), dextrose (20 g/L), and Bacto agar (20 g/L) in 1 L of (deionized or distilled) H2O, and autoclave. If necessary, add filter-sterilized acetamide to a final concentration of 600 mg/L (Solis-Escalante et al. 2013) or filter-sterilized cytosine to a final concentration of 1 mm (Hartzog et al. 2005) after autoclaving. After the medium has cooled to 55°C–60°C, pour into Petri dishes.
