P2 Buffer

  • 1. In a clean 1-L beaker, add:

    H2O, autoclaved, distilled 880 mL
    NaOH (10 m)  20 mL
    SDS (10%) 100 mL
  • 2. Add a sterile stir bar and stir for 5 min. To avoid SDS precipitation, add the NaOH solution to the H2O and stir for few minutes before adding the SDS solution. Label the bottle with the date.

    • P2 buffer should be made every 2 d and stored at room temperature to avoid SDS precipitation.

  • 3. If precipitation does occur, warm the bottle to 37°C and stir for a few minutes.
