Sonication Buffer
20 mm Tris-HCl (pH 8.0)
70 mm KCl
1 mm EDTA (pH 8.0)
10% glycerol
0.125% Nonidet P40 (10%)
Combine the reagents listed above. Immediately before use, cool buffer on ice and add 1 m DTT (5 mm final concentration) and protease inhibitor cocktail (PIC) (25×) (1× final concentration).
