Homogenization Buffer for Yeast

Reagent Quantity (for 1 L) Final concentration
Sucrose  113 g  0.33 m
Tris-Cl (1 m, pH 8.0) 300 mL  0.3 m
EDTA (0.4 m, pH 8.0)  2.5 mL  1 mm
EGTA (0.4 m, pH 8.0)  2.5 mL  1 mm
Dithiothreitol (DTT)  0.31 g  2 mm
6-Aminohexanoic acid (EACA)  13.1 g 100 mm
Store for up to 1 wk at 4°C; if stored longer, add fresh DTT before use.
