Paraformaldehyde (4%, w/v) in PBS

  • 1. In a fume hood, weigh and dissolve paraformaldehyde powder (Nacalai Tesque 26126-54) to prepare a 4% (w/v) formaldehyde solution in Phosphate-buffered saline (PBS). Prepare slightly less than the final required volume.

  • 2. Heat the solution to 60°C on a hot plate using a magnetic stirrer until completely dissolved.

  • 3. Adjust the pH of this solution to 7.4 using dilute (1 m) HCl. Top up the solution to the final volume after pH adjustment with PBS.

    • For experimental ease, it is possible to use stored frozen aliquots. However, using fresh 4% PFA is optimal for the best results.
