Paraformaldehyde (4%, w/v) in PBS

1. In a fume hood, weigh and dissolve paraformaldehyde powder (Nacalai Tesque 26126-54) to prepare a 4% (w/v) formaldehyde solution in Phosphate-buffered saline (PBS). Prepare slightly less than the final required volume.

2. Heat the solution to 60°C on a hot plate using a magnetic stirrer until completely dissolved.

3. Adjust the pH of this solution to 7.4 using dilute (1 m) HCl. Top up the solution to the final volume after pH adjustment with PBS.

For experimental ease, it is possible to use stored frozen aliquots. However, using fresh 4% PFA is optimal for the best results.
