SB Medium
In a large, sterile 4- to 5-L Erlenmeyer flask, add the following ingredients: 30 g of MOPS [3(N-Morpholino) propane sulfonic acid] and 90 g of tryptone. In a ventilated hood, add 60 g of yeast extract. Add 2 L of double-distilled (dd) H2O and mix on a magnetic stir plate while keeping covered.
Once the mixture goes into solution, add the remaining 1 L of ddH2O and let stir for another 3 min. Place the pH probe into the flask and titrate the media with either HCl or NaOH to attain a pH of 7. Together, this makes 3 L. Divide the contents among four individual 1-L flasks. Sterilize by autoclaving at 15 psi on a liquid cycle for 20 min at 121°C.
