XB (Extract Buffer)

Reagent Volume
Extract buffer (XB) salts (20×) 50 mL
Sucrose (2 m; filter-sterilized and stored in aliquots at −20°C) 25 mL
HEPES (K salt; 1 m, pH 7.7; filter-sterilized and stored in aliquots at −20°C) 10 mL
Combine and bring to 1 L with MilliQ H2O. Adjust pH to 7.7 with KOH. Make fresh on day of preparation, and store at 16°C until use.
