Hybridization Buffer for Colorimetric In Situ Hybridization

Reagent Quantity (for 3 L) Final concentration
Formamide 1.5 L 50%
Sodium chloride-sodium citrate buffer (SSC; 20×) 750 mL 5×
Torula RNA Type IX (Sigma-Aldrich R3629) 3 g 1 mg/mL
Heparin (Sigma-Aldrich H3393) 0.3 g 100 µg/mL
Denhardt's solution (100×) 30 mL 1×
Tween 20 3 mL 0.1%
CHAPS (Sigma-Aldrich C3023) 3 g 0.1%
EDTA 11.167 g 10 mm
Adjust to 3 L with H2O, check that pH is ∼7.5, and store at −20°C.
