Phosphate-Buffered Saline with Tween and BSA (PBSTB)

  • 1. Combine the following salt components in distilled H2O.

Reagent Final concentration
NaCl 150 mm
Na2HPO4 8 mm
NaH2PO4 2 mm
  • 2. Adjust salt components and distilled H2O to reach pH 7.4 by using NaOH or HCl for a final volume of 1 L.

  • 3. Autoclave the solution at 15 PSI for 20 min at 121°C on a liquid cycle. Verify the pH again.

  • 4. Add Tween 20 and dissolve BSA to final concentrations of 0.1% (w/v) and 5% (w/v), respectively. Adjust the volume to 1 L and sterilize with a 0.2-µm filter. Prepare fresh before use. Keep at 4°C.

    • Different concentrations of BSA (1%–5%, w/v) are used.

  • 5. When required, dilute PBSTB (5% BSA) with PBST to prepare solutions with lower concentrations of BSA.
